美國的一個小鎮,Jefferson Smith(James Stewart飾)是當地的童子軍的首領,深受青少年們的喜愛,被選為新的參議員,來到了華盛頓。遇到了他父親的老朋友,同為參議員的Joseph Paine(Claude Rains飾),並且迷上了Joseph的漂亮女兒。單純的Jefferson從來沒來過華盛頓,在華盛頓鬧了不少笑話,被媒體紛紛取笑。Joseph讓Jefferson起草一個提案,並讓他的女秘書Clarissa Saunders(Jean Arthur 飾)協助Jefferson。然而Jefferson卻發現,他的這個所謂參議員不過是其他老謀深算的參議員手中的木偶,他發現了他們一個巨大的陰謀。Jefferson決定以自己單薄的力量來對抗那群人,來改變這個腐敗的政治環境。到底他能否成功?史密斯先生的華盛頓之旅會如何結束?
