약대 졸업식날 마약 제조 혐의로 약사 자격을 뺏긴 맥클로이는 30년 후 마약 제조자가 된다. 리자드의 수하에서 코카인보다 51배 강하고, 엑스터시보다 51배 자극적인 신소재 마약 POS-51D을 제조한 맥클로이는 세계 최고의 마약상 듀란과 거래를 위해 또 다른 음모를 꾸민다. 하지만 거래하는 날 듀란과 맥클로이는 나타나지 않고 약속장소는 폭파된다. 가까스로 목숨을 건진 리자드는 맥클로이의 배신에 분노해 미모의 킬러 다코타에게 맥클로이 제거를 명령한다.
